Abstract: Objectives To elucidate the function and underlying mechanism of lncRNA(long non-coding RNA) SRGAP3-AS2(SRGAP3 antisense RNA 2) on the proliferation,migration and invasion of non-small cell lung cancer(NSCLC).Methods The expression of SRGAP3-AS2 in NSCLC tissues and H23 cells was detected,and the relationship between the expression of SRGAP3-AS2 and the clinicopathology of NSCLC was analyzed.H23 cells were divided into the NC group,pc-SRGAP3-AS2 group and pc-SRGAP3-AS2+JAK2 activator group.Cell proliferation,migration,invasion,MMP-2,Bcl-2,p-JAK2 and p-STAT3 expressions were detected in each group.Results The expression of SRGAP3-AS2 was lower in NSCLC tissues,which was related to lymph node metastasis and TNM stage(P<0.05).Compared with the NC group,the cell viability,the wound healing rate,the number of invasive cells,the expressions of MMP-2,Bcl-2,p-JAK2 and p-STAT3 in the pc-SRGAP3-AS2 group were decreased(P<0.05).Compared with the pc-SRGAP3-AS2 group,the cell viability,wound healing rate,number of invasive cells,MMP-2,Bcl-2,p-JAK2 and p-STAT3 expression in the pc-SRGAP3-AS2+JAK2 activator group increased(P<0.05).Conclusion The lower expression of SRGAP3-AS2 in NSCLC tissue is related to lymph node metastasis and TNM.Overexpression of SRGAP3-AS2 can inhibit JAK2/STAT3 signaling and inhibit the proliferation,migration and invasion of NSCLC cells H23.
